Tipsarevic, 31, played just 13 competitive singles matches last year after returning, with his last appearance at the US Open.
A former world number eight, he has slipped to 410th in the world as a result of his inactivity. He was expected to take his place at Melbourne Park with a protected ranking, but his withdrawal was confirmed on Wednesday.
